结晶案例:乙腈/DMF体系加水析出油怎么办?再加点DMF

DMF为溶剂做工艺,常规思路都是能少加DMF则少加,主要理由是DMF后处理很麻烦,还需考虑残留问题,其次理由才是回收、环境因素。

对于结晶来讲,什么条件都可以尝试,什么溶剂都可能用到。
小编经验案例,反应采用DMF,后处理加水析晶,产品过滤困难,解决方案是体系先加乙醇再滴加反溶剂水。

  • 酸胺缩合反应,乙腈为溶剂,T3P的DMF溶液为缩合剂,反应后处理,加水结晶,基本得到油。
  • 解决方案,反应体系再加入一定量DMF,再滴加水。

  • 角色互换,如果是小编,面对成油的问题,会先浓缩去除乙腈,然后走萃取策略,再结晶。也许会尝试其他结晶策略,但是很难会加入一定量DMF,这就是工艺。

实验操作
An inert reactor was charged with 14 (87.4 kg, 1.00 equiv), 12 (59 kg, 1.05 equiv), MeCN (392 kg, 500 L), and NEt3 (29 kg, 1.5 equiv). A 50% T3P in DMF (183 kg, 1.5 equiv) was slowly charged maintaining an internal temperature of no more than 35 °C. The mixture was agitated at 20 °C for 3 h, and then DMF (87 kg, 92 L), water (218 kg), and 15 (0.26 kg) were charged. To the mixture was added water (392 kg) over no less than 2 h. The resulting slurry was aged for 2 h and then filtered. The filter cake was washed with a solution of water (392 kg) and MeCN (305 kg, 388 L), and then the solids were dried under vacuum at 50 °C to afford 15 as a white crystalline solid (129 kg, 91.5% yield, 100% purity by UPLC analysis)
