实验操作An inert reactor preconditioned with MeCN was charged with 13·NADL (140 kg),
Pd(PPh3)2Cl2 (2.66 kg, 0.03 equiv), and 5 (40.6 kg, 1.05
equiv). The reactor was inerted and then charged with MeCN
(420 kg, 534 L) and NEt3 (126 kg, 174 L, 5.0 equiv). The
mixture was agitated at 70 °C for 6 h, water (70 kg) was
charged, and then the mixture was cooled to 50 °C. NAC (4.2
kg, 2.8 kg/kg) was charged, and the contents were agitated for
6 h at 50 °C. The contents were then cooled to 22 °C over 2 h
and agitated at 22 °C for 2 h. To the reactor was charged water
(1050 kg) over no fewer than 2 h. The resulting slurry was
aged at 22 °C for 4 h and then filtered. The filter cake was
washed with a solution of water (364 kg) and MeCN (140 kg,
178 L), and then the solids were dried under vacuum at 50 °C
to yield 14 as a white crystalline solid (100 kg, 88.3% yield,
100% purity by UPLC analysis, 121 ppm Pd by ICP-MS)
