最初合成路线如下
  • 化合物3经偶联反应获得化合物4,然后经迈克尔加成得到化合物5,不对称还原羰基得到化合物6,水解、关环、α-位烷基化得到目标物

改进路线
  • 采用傅克酰基化法得到化合物4后,路线和最初路线类似,只是不对称还原的底物发生了变化,改用异丙酯底物11代替最初路线的化合物5
  • 有意思的是酯交换步骤,甲酯在浓硫酸条件和异丙醇存在下,制备异丙酯。

To a 2500 L enameled  reactor equipped with a NaOH scrubber were charged a solution of 5 in isopropanol from the previous step (666.6 kg of solution, 32% w/w, l.0 equiv based on HPLC assay) and isopropanol (860 L, 3.6 vol). Sulfuric acid (159.3 kg, 2.4 equiv, 86 wt % solution) was added over 15 min, and the mixture was heated to 80−84 °C for 15 h. Isopropanol was distilled at 85 °C and atmospheric pressure over 9 h and replaced with fresh isopropanol (800 L, 3.4 vol). After conversion to the isopropyl ester, the mixture was concentrated by distillation at atmospheric pressure over 9.5 h until the total volume reached
640 L.
