近日,结构药理学与中药化学生物学研究所(实验室)在国际知名期刊Phytochemistry上发表了题为”Non-alkaloid components with inhibitory activity against LPS induced NO
production in RAW 264.7 cells isolated from the roots of Sophora flavescens”的研究文章(doi: 10.1016/j.phytochem.2024.114288)。沈阳药科大学无涯创新学院博士生徐阳为本文第一作者,沈阳药科大学无涯创新学院陈丽霞教授和福建中医药大学药学院李华教授为共同通讯作者。
苦参(Sophora
flavescens)是豆科(Leguminosae)槐属(Sophora)植物,主要分布在亚洲中西部,以根为主要药用部分。作为一种中药,其主要用于治疗湿疹、血便、皮肤瘙痒等症状。苦参中的化学成分主要分为生物碱类和非生物碱类成分,以往对苦参中的的研究重点集中在其色戒类成分,对其非生物碱类成分的研究较少。从苦参中分离到的非生物碱类成分包括黄酮类、三萜类以及木酚素类等,其中黄酮类是其非生物碱的主要成分。
本文对苦参干燥根部进行研究,分离得到49个化合物,其中包括3个新的黄酮,5个新的异戊烯黄酮类以及41个已知化合物。根据其理化性质和波谱学手段(NMR、HR-ESI-MS、IR,ECD等)鉴定了这些化合物的结构。此外,评估了所有化合物抑制脂多糖(LPS)诱导的小鼠巨噬细胞RAW264.7释放NO的活性,其中化合物11和22具有较好的抑制NO释放活性,IC50值分别为4.58 ± 0.66 and 4.53 ± 0.66 μM。
