RNA-directed DNA methylation (RdDM)是调控植物基因表达的一种重要机制。本研究从突变体筛选入手,发现一个KOW DOMAIN-CONTAINING TRANSCRIPTION FACTOR1 (KTF1)(an essential component of the RdDM pathway)的突变体呈现对盐胁迫敏感表型,分析显示该突变体中的ZmKTF1的启动子发生了 insertion–deletion mutations (InDels).
Whole-genome bisulfite sequencing显示,玉米的ZmKTF1突变会显著降低在整个基因组范围内CHH (H = A, C, or T) 位点的甲基化水平,从而调控了氧化还原酶相关基因表达,导致植物体内活性氧水平升高。
在生理水平上,ZmKTF1还影响了植物体内Na+水平。
