Alex's research group aims to identify potential weaknesses in the otherwise robust signalling networks of cancer cells which can be exploited for therapeutic intervention. Specifically focusing on the initiating events in cancer establishment as well as the events triggering the spread of cancer throughout the body. Commonly using mass spectrometry methods, such as interaction, expression, exosome/secretion and PTM proteomics to initially generate signalling networks. Subsequently, his group use this integrated information to generate new and testable hypotheses. This unbiased data which we use in combination with mathematical modelling can explain unintuitive cellular behaviours.

His group is now applying these methods developed in pre-clinical models to investigate large cohorts of tumour tissues with the aim to develop accurate and sensitive early-detection methods, predict response to disease and to develop novel therapeutics. Currently, he is focusing on how to increase tumour immunogenicity by augmenting Innate Immune signalling and is developing a novel biomarker panel for Ovarian cancer.

Talk abstract

Recent advances in spatial and single-cell (sc) transcriptomics have transformed our understanding of disease mechanisms and tissue homeostasis. Single-cell analysis has provided unprecedented insights into cellular heterogeneity, revealing the complexity of tissue microenvironments and how individual cells contribute to both health and disease.
Very recently, improvements to software and hardware have made proteomics at the SC level achievable. In this presentation, we will explore how cutting-edge techniques in sc proteomics, powered by advanced MS technology, are reshaping cancer research, providing deeper insights into tumour heterogeneity. 
Furthermore, we will discuss the application of spatial proteomics, particularly from formalin-fixed, paraffin-embedded (FFPE) tissues, which allows for the retrospective analysis of archival samples while maintaining histological context. These approaches are not only redefining our capacity to study the molecular landscape of cancer but also offer new potential for personalized medicine and targeted therapies.
近年来,空间和单细胞(single cell)转录组学的进展极大地改变了我们对疾病机制和组织稳态的认识。单细胞分析为细胞异质性提供了前所未有的洞察,揭示了组织微环境的复杂性,以及单个细胞在健康和疾病中的作用。
最近,软硬件的升级使得在单细胞层面开展蛋白质组学研究成为现实。在本次报告中,我们将探讨先进质谱技术推动的单细胞蛋白质组学前沿技术如何重塑癌症研究,并加深我们对肿瘤异质性的理解。
此外,我们还将讨论空间蛋白质组学的应用,特别是在福尔马林固定、石蜡包埋(FFPE)组织中的应用,它使我们能够在保持组织结构的前提下,对存档样本进行回顾性分析。这些方法不仅重新定义了我们研究癌症分子图谱的能力,还为个性化医疗和靶向治疗带来了新的可能性。
