酮戊二酸(KGA)和甲烷三乙酸(MTA)是一类重要的脂肪族多元羧酸,被广泛应用于合成膳食补剂、热稳定涂料、可生物降解弹性体和金属有机框架材料等。然而,它们的制备过程存在着效率低、能耗高和污染严重等缺点(图1a)。因此,开发高效可持续的合成方法至关重要。

近日,北京大学刘海超教授团队发展了一种从生物质衍生的丙酮酸和乙醛酸一锅法绿色且高效合成酮戊二酸和甲烷三乙酸的新方法(图1b)。该方法中使用的丙酮酸、乙醛酸的分子结构与酮戊二酸、甲烷三乙酸在断裂C3-C4键后形成的片段结构相似。同时,它们均可通过生物质基乳酸和乙醇酸的选择性氧化制备。因此,与传统的合成方法相比,以生物质基丙酮酸和乙醛酸为原料,通过连续的交叉羟醛缩合和加氢脱氧反应合成酮戊二酸和甲烷三乙酸具有明显的优势。
2. 酮戊二酸(KGA)的可持续合成
具体而言,丙酮酸和乙醛酸首先通过羟醛缩合反应转化为2-氧代-4-羟基戊二酸中间体,然后该中间体发生连续的脱水和加氢反应,实现酮戊二酸的可持续合成(图2)。TiO2负载的Pd催化剂在该反应中表现出较高的效率和稳定性,在100 ℃和1.0 MPa H2的反应条件下,酮戊二酸的收率达到85%。

进一步,酮戊二酸和乙醛酸发生缩合反应转化为3-羧甲基-2-羟基-4-氧代戊二酸中间体,然后该中间体发生氢解反应转化为甲烷三乙酸(图3)。机械混合的Pd/TiO2和MoOx/TiO2催化剂在该反应中表现出较高的氢解效率和稳定性,在200 ℃和2.0 MPa H2的反应条件下,甲烷三乙酸的收率达到86%。其中,分散的MoOx物种主要负责氢解C-OH键,金属Pd主要负责活化氢气、加氢中间体中C=C和C=O键。
