Cancer and diabetes: cell signaling mechanisms

Various cancers and diabetes mellitus are highly prevalent diseases associated with altered cell signaling mechanisms. Epidemiological data shows a 2-fold increase for developing cancers such as liver, pancreas, and endometrial cancers in patients with diabetes mellitus. There is also an increased risk (~1.4 fold) for developing breast, bladder, and colon cancers in patients with type 2 diabetes mellitus (T2DM). However, a lower risk of developing prostate cancer is found in men with diabetes mellitus than in men without this disease. The epidemiological association between an increased risk of developing T2DM has also been reported in patients with or without treatment for cancer.
This special issue (SI) focuses on the molecular mechanisms that could explain these findings emphasizing potential cell metabolic changes found in the pair cancer/diabetes mellitus. Although there are several known cell metabolic alterations in patients with T2DM and cancer, most show altered unfolded protein response (UPR-response) in the endoplasmic reticulum and changes in mitochondrial activity leading to increased reactive oxygen species (ROS) production and intracellular calcium levels. The contributions to this SI will cover how these metabolic alterations impact cell signaling involved in the modulation of biological membranes, ion channels, membrane transporters and receptors.
The potential interdependence of cancer and diabetes mellitus are common diseases that increase in incidence with age. Unveiling mechanisms that might link these two metabolic diseases are relevant for diagnostic, prognostic, and therapeutic purposes in patients with cancer, diabetes mellitus, or both.
